چکیده
Abstract Extensive decoration of backbones is a major factor resulting in resistance enzymatic conversion hemicellulose and other branched polysaccharides. Employing debranching enzymes the main strategy to overcome this kind recalcitrance at present. A carbohydrate-binding module (CBM) contiguous amino acid sequence that can promote binding various carbohydrates, thereby facilitating hydrolysis. According previous studies, CBMs be classified into four types based on their preference ligand type, where Type III IV prefer polysaccharides than linear thus are able specifically enhance hydrolysis substrates containing side chains. With role dominating substrates, could represent non-catalytic approach overcoming side-chain recalcitrance.

The Contribution of Non-catalytic Carbohydrate Binding Modules to the Activity of Lytic Polysaccharide Monooxygenases.
Lignocellulosic biomass is a sustainable industrial substrate. Copper-dependent lytic polysaccharide monooxygenases (LPMOs) contribute to the degradation of lignocellulose and increase the efficiency of biofuel production. LPMOs can contain non-catalytic carbohydrate binding modules (CBMs), but their role in the activity of these enzymes is poorly understood. Here we explored the importance of ...

The enzymic degradation of insoluble polysaccharides is one of the most important reactions on earth. Despite this, glycoside hydrolases attack such polysaccharides relatively inefficiently as their target glycosidic bonds are often inaccessible to the active site of the appropriate enzymes. In order to overcome these problems, many of the glycoside hydrolases that utilize insoluble substrates ...
متن کاملThe interaction of carbohydrate-binding modules with insoluble non-crystalline cellulose is enthalpically driven.
Natural cellulose exists as a composite of cellulose forms, which can be broadly characterized as crystalline or non-crystalline. The recognition of both of these forms of cellulose by the CBMs (carbohydrate-binding modules) of microbial glycoside hydrolases is important for the efficient natural and biotechnological conversion of cellulosic biomass. The category of CBM that binds insoluble non...
متن کاملFunctional studies and engineering of family 1 carbohydrate-binding modules
The family 1 cellulose-binding modules (CBM1) form a group of small, stable carbohydratebinding proteins. These modules are essential for fungal cellulose degradation. This thesis describes both functional studies of the CBM1s as well as protein engineering of the modules for several objectives. The characteristics and specificity of CBM1s from the Trichoderma reesei Cel7A and Cel6A, along with...

Competition isotherms are used to identify the set of cellulose substructures to which cellulose binding modules (CBMs) from families 2a, 3, 4, 9, and 17 bind. The experiments are based on coupling a unique fluorescent tag to each CBM in a manner that does not alter the natural binding properties of the CBM and therefore allows the surface and solution concentrations of each CBM to be monitored...
